There's always the cotton rope trick. Find a piston that's approaching
TDC when the crank is rotated backwards. Make sure the valves are
CLOSED on that cylinder. Remove the spark plug and feed a length of
cotton rope into the cylinder so that the piston is stopped by the rope
when the crank bolt is turned counterclockwise. This should provide
adequate resistance to undo the bolt without placing undue stress or
impact on the piston or anything else.

Out of the car offers a wide range of methods. Personally, I'd just use
my
impact wrench. But I have a feeling you don't have a pneumatic system. I
don't know the orientation of the engine, but a friend holding a large
screwdriver in the ring gear braced against the block while you use a
socket
and long breaker bar will work nicely. If you are marooned on a desert
island, a sharp blow or two to the handle of the breaker bar
(anti-clockwise) with a large mallet would probably do it. Although
your
engine is not a turbo, I'm pretty sure it is a 3S-GE and will most
likely
need both belt pensioners and the water pump replaced. Don't forget new
O-rings for the water pump. The procedure for tensioning the belt is on
that web site I sent.
Cleanliness is next to Godliness here. Are you beginning to wish you'd
purchased an engine from the breakers yet?
